What the Gut-Brain Axis Actually Is
The gut-brain axis is a two-way communication system connecting your gastrointestinal tract to your central nervous system. It travels through the vagus nerve, through the immune system, through hormones, and through molecules produced by the trillions of microbes that live in your gut.
A few numbers help paint the picture. Your gut contains roughly 100 trillion microorganisms, more than the number of cells in your body. It produces about 95 percent of the body's serotonin, a primary neurotransmitter involved in mood, sleep, and appetite. And through the vagus nerve, gut signals influence brain regions responsible for stress response, memory, and emotional regulation.

Where Functional Mushrooms Fit In
Functional mushrooms contain a class of compounds called beta-glucans, along with other polysaccharides that act as prebiotics, food for the beneficial bacteria in your gut. This is one of the most interesting findings in recent mushroom research. Some of the cognitive and mood benefits people attribute to functional mushrooms may actually begin in the gut, not the brain.
One of the most illuminating studies on this connection was published in Scientific Reports in 2021. Yao et al. showed that reishi (Ganoderma lucidum) promoted sleep in mice through a gut microbiota-dependent pathway that involved serotonin signaling. In other words, the calming, sleep-supporting effects of reishi appear to travel from the gut, up through the vagus nerve and neurotransmitter systems, to the brain. This was a real shift in how researchers had been thinking about this mushroom's workings.
Lion's Mane, Beyond the Brain
Lion's mane has been most famous for its effects on nerve growth factor and cognition. But newer research is expanding the picture. A 2024 study on Hericium erinaceus in GeroScience / Nutrients (referenced in the Alzheimer's Drug Discovery Foundation's Cognitive Vitality review) found that lion's mane supplementation was associated with meaningful changes in gut microbiome composition, including increases in beneficial short-chain fatty acid-producing bacteria like Bifidobacterium and Faecalibacterium prausnitzii. Short-chain fatty acids are one of the primary ways gut bacteria influence brain and immune function.
This changes how we think about a lion's mane routine. Cognitive benefits may appear faster in some people due to direct effects on brain signaling. But the deeper, more durable benefits may build over weeks and months as the gut microbiome shifts. That's part of why lion's mane is best taken consistently, not just when you're preparing for a big day.
Reishi, Cordyceps & Turkey Tail
Reishi's sleep and stress support, as we've discussed, appears to travel through the gut. But reishi also has a long-studied effect on immune modulation, and much of that occurs in the gut, where about 70 percent of your immune system lives.
Cordyceps supports cellular energy production and physical endurance, and while less research has focused on the microbiome specifically, its polysaccharides are also prebiotics. Turkey tail is one of the most-researched functional mushrooms for gut health, with its polysaccharide-K (PSK) and polysaccharide peptide (PSP) compounds having a substantial evidence base supporting immune and gut microbiome support.
Taken together, this points to something important: the effects of functional mushrooms don't fit neatly into single categories. Cognitive, immune, stress, and gut effects are all interwoven, because in your body, they are.
What This Means for a Practical Wellness Routine
If you're new to functional mushrooms and specifically interested in the gut-brain angle, a few principles can help. First, consistency matters more than dose. Microbiome shifts happen over weeks, not days. Give any mushroom protocol at least six to eight weeks before drawing conclusions.
Second, pair mushrooms with fiber-rich foods. Beta-glucans work synergistically with other prebiotic fibers. If your diet is low in vegetables, fruit, and whole grains, even the best mushroom blend has less to work with.
Third, notice what you notice. Improvements in sleep, mood, digestion, immune resilience, and cognition can all be signals that the gut-brain conversation is running more smoothly. Keep an informal log if it helps you see what's shifting.
